Purpose:To assess the safety of Nexium 20 mg and Esomeprazole 20 mg capsules administered in adults.

Materials and methods:An open-label, open-label, observational study was conducted at the Department of Clinical Endocrinology of University of Krakóna Medical Center, Krakól, Krakól, Krakól, Poland. All patients with mild to moderate esomeprazole toxicity with suspected proton pump inhibitor (PPI) therapy were randomly assigned to receive either 20 mg or 40 mg of Nexium 20 mg, the active ingredients of the medication, for 12 weeks. In the 20 mg group, 5 patients (3.4%) experienced gastrointestinal adverse events compared with 2 patients (1.6%) in the 40 mg group (P = 0.07). No significant differences were found in the frequency of gastric irritation, peristalsis, or esophagitis in the 20 mg group. In the 40 mg group, the most common adverse events (39.2%) were headache, nausea, abdominal pain, and flatulence, compared with the 20 mg group (35.7%) (P = 0.09). There were no significant differences in the frequency of gastrointestinal hemorrhage, peristalsis, or esophagitis in the 20 mg and 40 mg groups.

Conclusion:Esomeprazole 20 mg was more effective than 20 mg in the treatment of esomeprazole-associated gastrointestinal adverse events.
